     บทคัดย่อ Background Department of Livestock Development has developed new synthetic breed from crossbred pigs between Largewhite and Pietrain, so call “Pakchong III” for promoting as an alternative terminal sire in finishing pig program in Thailand. To develop new synthetic breed, crossing between two purebreds follow by inter se mating for 5 generations is a regular method to guarantee the theoretical homozygosity for new breed over 95%. However, the homozygosity in genome level might not assure. There are several tools for genetic characterization. Microsatellites are recommended by FAO for the characterization of genetic distances between breeds. Benefit of their codominant and multiallelic nature prove to be efficient in genetic diversity. Multivariate procedures using Principal Component Analysis (PCA) is found very useful for genetic studies concerning breeds or populations from the same species. Regardless of prior grouping, genetic relationships among individuals are obtained by condensing information from several alleles of different loci from genome into a few variables called Principal Component Variables (PCV). The purpose of this study was to investigate genetic characterization of synthetic pig population from purebred population using PCV plot from microsatellite data. Result Blood samples were collected from 252 pigs of 54 Pietrain (PT), 76 Large White (LW), and 122 PTxLW pigs (PC3) to isolate genomic DNA and genotyped using 14 loci of microsatellites. Nei's genetic distances between animals were calculated from 52 alleles and transformed to PCV. Plotting the first two PCV showed the increasing of homozygosity in synthetic population from the first to fourth year of selection. The PCV boundary of PC3 was clearly overlapped with PT and LW in the first generation (F1), which revealed strong degree of heterozygosity. However, the overlapping pattern was decreased, which showed the increasing of homozygosity during the inter se mating and selection for each generation. The PCV boundary of synthetic pigs was almost separated from purebred populations in the fourth year of selection. The complete discrimination of PC3 from LW, PT was expected within the next few year of selection. Conclusions It was concluded that principal component plot from microsatellite data could be used as an efficient method for breed characterization.
