| บทคัดย่อ |
A simple method for extraction of inulin from Jerusalem artichoke tubers by accelerated solvent extraction (ASE) was developed using water as the extractant under high pressure condition. The parameters affecting the extraction included temperature and time were investigated. The studied temperatures and times were 70-100๐C and 10-30 min, respectively. The quantity of inulin was determined based on the contents of glucose and fructose which were spectrophotometrically detected via periodate reaction. The degree of polymerization (DP) of inulin was also calculated based on glucose and fructose contents. The optimum condition for the extraction were 80๐C and extraction time of 20 min at 1500 psi. The inulin content obtained from 4 varieties of Jerusalem artichoke were in the range of 64 to 70% dry weight and DP was in the range of 14.4-19.7. It was found that longer extraction time could extract high DP inulin. In addition, the inulin profiles under different extraction conditions were monitored and evaluated using high performance anion exchange chromatography coupled with pulsed amperometric detection (HPAEC-PAD). |
