     บทคัดย่อ Objective: Cigarette smoking is related to oxidative stress and contributes to the risk of metabolic syndrome (MetS). Two polymorphisms of PON1, Gln192Arg (A575G; rs662) and C-108T (rs705379), alter its enzyme ability to protect low density lipoprotein (LDL) from oxidation. Although the associations between these polymorphisms and the MetS components have been reported, they may be modified by smoking. The objective of this study was to investigate the association of PON1 gene polymorphisms (rs662 and rs705379) with the risk of MetS in Thai cigarette smokers. Method: A total of 212 Thai cigarette smokers (mean age: 54.4±10.0 years; 54 former and 158 current smokers) were recruited. The prevalence of MetS was determined according to modified National Cholesterol Education Program III criteria. PON1 gene polymorphisms (rs662 and rs705379) were determined in genomic DNA. Result: The respective frequencies of rs662 A and G alleles were 30.9% and 69.1%, whereas the C and T alleles of rs705379 were 45.8% and 54.2%. Only rs705379 revealed the relationship with MetS; TT was associated with an increased risk of MetS (OR = 3.10, 95%CI 1.18-8.16) and with higher waist/hip ratio, as compared with CC. For rs662, the subjects with GG had significantly lower levels of total cholesterols and LDL-C (p=0.032 and 0.043), but had higher systolic blood pressure (p=0.036) than those with AA genotype. Weak linkage disequilibrium (D'=0.72) between these two polymorphisms was observed. Conclusion: The present study demonstrated the association of PON1 gene polymorphism, rs705379, with the risk of MetS in Thai subjects with cigarette smoking.
