บทคัดย่อ The highest co-prevalence of opisthorchiasis caused by Opisthorchis viverrini infection and diabetes mellitus (DM) is found in northeastern Thailand. However, the association between DM and O. viverrini infection is unclear. Moreover, the effect of parasitic infection on DM is controversy. The aim of this study was to investigate the association of O. viverrini infection and the progression/severity of type 1 diabetes induced by streptozotocin (STZ) in hamsters. The experimental study was carried out on four groups of hamsters (n= 5 per group): (1) normal control, (2) O. viverrini-infected, (3) STZ-induced and (4) STZ-induced and followed by O. viverrini-infected at 2 months post-treatment. All hamsters were sacrificed at 3 months post-treatment. The results revealed that O. viverrini infection in STZ-induced diabetes was significantly higher of liver per body weight ratio than in other groups. O. viverrini infection synergistic increased of both liver cell and bile duct damages in STZ-induced diabetic hamsters as evaluated by liver function test and TUNEL assays. Furthermore, O. viverrini infection in STZ-induced diabetic hamsters significantly increased liver cell hypertrophy and lipid accumulation, bile canaliculi alteration, hepatocyte and bile duct proliferation compared to other groups. Moreover, globet cell metaplasia was found only in STZ-induced diabetes and infected with O. viverrini. Additionally, the size of liver fluke in STZ-induced diabetes was smaller than in O. viverrini-infected normal hamsters. Indeed, these results suggest that O. viverrini infection synergistic increases on the hepatocyte and bile duct abnormalities in diabetes condition. The study may have implication for chemoprevention and therapy approach of DM patients-infected with O. viverrini.
