     บทคัดย่อ N-Nitrosodiethanolamine (NDELA) is one of the most common nitrosamines found in a wide variety of preserved food and beverages which is activated within the body to form a carcinogen leading to tumors/cancers. Hepatic CYP2A6, CYP2A13, and CYP2E1 are considered to be of particular importances for biotransformation of NDELA. Constitutive androstane receptor (CAR) is a nuclear receptor responsible for metabolism of xenobiotics and regulation of CYP2A6, CYP2A13 and CYP2E1 expression in the liver. The study aimed to evaluate the effects of NDELA on the expressions of CYP2A6, CYP2A13, CYP2E1, and CAR mRNA in human liver hepatoma derived cell line (HepG2). HepG2 cells (5×105 cells in a 6-well plate) were treated with NDELA (1μM, 10μM, and 100μM) for 24 h. Total RNA was isolated using guanidinium-thiocyanate-phenol-chloroform method and cDNA was reverse-transcribed. The mRNA expression was analyzed by qPCR using specific primers and SYBR green, compared to that of GAPDH (a reference gene). The highest concentration of NDELA (100 μM) significantly up-regulated the expression of CYP2A6 mRNA for 3-fold. Besides, all concentrations of NDELA induced the expression of CYP2A13 mRNA in a concentration-dependent manner with the maximum induction of 110-fold. Similarly, all concentrations of NDELA significantly elevated the mRNA level of CYP2E1. The highest concentration of NDELA (100 μM) noted a significant induction on the CAR expression. Therefore, the regulation of CYP2A6, CYP2A13 and CYP2E1 expression by NDELA might involve with CAR, at least in part.
