     Abstract Two-dimensional gel electrophoresis (2-DE) technique is wildly used for protein research. The objective of this work was to to use 2-DE coupled with analysis of gels for identification of proteins differentially expressed in response to water deficit stress. Sugarcane plants were planted and water stress imposed. Soil moisture content (SMC), Leaf water potential (LWP) and relative water content (RWC) indicated that sugarcane plants were under water deficit stress. Proteins were extracted from sugarcane leaves under water stress and adequate water supply and isolated in three replicated gels using 2-DE. Proteins were visualized by silver nitrate staining and the gels analyzed using Melanie 7 software. There were 9 down-regulated and 24 up- regulated proteins that displayed significant changes (p≤0.1-.05) in response to water deficit stress. Identification of the responsive proteins may provide insight into cellular mechanisms of drought tolerance in sugarcane.
