Abstract Lawsone (2-hydroxy-1,4-naphthoquinone), a red-orange dye presented in leaves of henna plant (Lawsonia inermis), used for cosmetic purposes for skin, hair, and fingernails. This study was aimed to monitor dynamic events of human epidermal carcinoma keratinocytes (A431), when exposed to a series of concentration (1.5 M – 15 mM) of Lawson, based on cell growth and proliferation assay using the xCELLIgence system. The system monitored cell response profile using electrical impedance by measuring real-time cell index. After cell suspensions were seeding into the E-plate 96 well plate (3000 cells/well), A431 were monitored every 15 min until log phase. IC50 from the dose-response curves was found to be about 0.5 mM. This was correlated to MTT end point assay. Real-time and continuous monitoring of keratinocytes which could be suggested cells response mechanism based on dose dependent.
