     Abstract Introduction: Diagnosis of opisthorchiasis requires sensitive and specific methods for screening and assessment the infection. The purposes of the study were to compare and evaluate Formalin ethyl acetate concentration technique (FECT) and PCR by using a manual DNA extraction method for detection of Opisthorchis viverrini infection in hamster model. Methods: Hamsters were infected with 50 O. viverrini metacercariae/animal. Fecal samples were collected at weekly intervals from 0-12 wk and examined by a quantitative formalin-ethyl acetate concentration (FECT) and PCR. The manual CTAB method was used for DNA extraction and inhibitor removal from the fecal specimens for DNA detection by polymerase chain reaction (PCR). To examine the source of the fecal DNA, eggs or tegument of O. viverrini adult worms were added to clean feces and processed by CTAB method for PCR. Results: The assay could detect O. viverrini DNA at feces 3 weeks post-infection (80%), whereas the FECT techniques detected the eggs in the feces from 4 weeks post-infection onward. Moreover, in group of EPG negative hamsters, 25% were PCR positive. The results also showed that PCR were positive in specimens seeded with either worm tegument. Conclusions: The PCR-based diagnosis using a manual extraction method PCR is a potential tool for diagnosis opisthorchiasis and it detected DNA before egg recovery by a conventional method.
