     Abstract Monosodium glutamate (MSG) is a flavor enhancer commonly used to prepare various kinds of foods in modern life styles. MSG may be a factor that causes male infertility since it has been demonstrated the effects of high dose MSG on testicular weight, sperm production, and testosterone levels in experimental animals; except, its effects on the testicular proteomic level. Therefore, this study aims to investigate the effects of high dose MSG on both testicular morphology and protein phosphorylation involved in spermatogenesis and testosterone synthesis. Two groups (control and MSG treated) of adult male rats were gastric fed with distilled water or MSG solution (6 g/ kgBW) for consecutive 15 days. At the end of experiment, right testis in each group was observed gross morphology and histology, whereas the left side was used to study the total protein profiling using SDS-PAGE. Additionally, testicular protein lysate was determined for protein phosphorylation using immunowestern blotting technique. Present results showed the significant reduction of testicular size, weight, and diameter of seminiferous tubule in the testis of MSG group. Moreover, we found a 71 kDa protein band present in the testicular lysate of MSG group. In western blotting result, the obvious reduction of protein phosphorylation in MSG group was observed. To our knowledge, high dose MSG may cause male infertility via reduction of testicular protein phosphorylation, leading to testis dysfunctions. Therefore, toxicity of this issue must be concerned.
