Monosodium glutamate (MSG) is a flavor enhancer commonly used as a food additive
in the modern life style. Although the toxicity of MSG is still controversy, MSG could be a
factor that causes male infertility since it has been demonstrated its effects on animal testes
except other male reproductive organs. To verify the effects of MSG on entire male
reproductive system, this study investigated the effects of various dose MSG on organ
weights and morphologies of the testes, epididymis, vas deferens, seminal vesicle, and penis
in male rats. Additionally sperm concentration and status of sperm acrosome reaction (AR)
were also observed. In experimental design, thirty-two adult male Spruge-Dawley rats (180-
200 g) were divided into four groups (8 animals/group) and were gastric fed with distilled
water (control group) or non-acidic MSG solutions (0.25, 3, and 6 g/kg BW) two periods per
day for consecutive 30 days. The results showed no significant differences in the testicular
weight, morphology and sperm AR status in rats treated with MSG, compared to the control
group. In contrast, the epididymis plus vas deferens and seminal vesicle of rate received
MSG (6 g/kg BW) showed significant decrease in weights and morphologies as compared
with the control (P<0.05). Moreover, the sperm concentrations retrieved from the
epididymis and vas deferent in rats received MSG at only doses of 3 and 6 g/kg BW were
significantly decreased (P<0.05) as compared to the control or MSG-treated group (0.25 g
/kg BW). To our knowledge, this study demonstrated that high dose of MSG resulted in the
reduction of sperm concentration and hypotrophy of the seminal vesicle, leading to less
productions of seminal fluid containing mainly sperm nutrients. Possibly, this may a cause of
male infertility. Therefore the use of MSG at high concentration should be concerned. |
