Abstract |
Lentinus polychrous, Thai local edible mushroom, was cultured under solid state fermentation. The ethanolic extract of their mycelia (LPME) was investigated for its in vitro effects in immune-restoration. Balb/cMal mice were pre-treated with cyclophosphamide (Cyc) for four days prior to removal of their spleens for splenocyte cultivation. The cytotoxic effect of LPME against Cyc-treated splenocytes was determined. In addition, the effect of LPME on Cyc-treated splenocyte proliferation was evaluated in the presence of T-cell and B-cell inducing mitogens, phytohemagglutinin (PHA) and poke weed mitogen (PWM), respectively. The results showed that the Cyc treatment decreased size of spleen and viable cell numbers of splenocytes compared with Cyc untreatment. The LPME exhibited cytotoxicity against Cyc-treated splenocytes with IC50 of 76.12 ± 8.24 µg/ml. In the presence of PHA and PWM, the LPME induced the proliferation of the Cyc-treated cells significantly. These results indicate that the LPME can restore the proliferation of both immunosuppressed splenocytes. |
