Research Title Effect of Combined Tetrahydrocurcumin and Deferiprone Chelation Therapy on Iron Sucrose-Induced Iron Overload in Mice

     Abstract Iron overload is an excessive accumulation of iron in the body. This condition can result from inherited or acquired causes such as sickle cell disease and numerous blood transfusions especially in major thalassemia patients. Excessive iron has the potential to generate reactive oxygen species (ROS) via Fenton reaction, which leads to oxidative stress and subsequently damage of biomolecules as well as impairment of vascular function. In the present study, we demonstrated that administration of iron sucrose (10 mg/kg/day, i.p.) for 8 weeks to male ICR mice. Administration of iron sucrose induced iron overload as evidence by increasing of serum iron indices when compared with untreated-animals (p<0.05). Iron toxicity also exerted deleterious effects by raising arterial blood pressure which were related with increased vascular superoxide production, elevated plasma malondialdehyde (MDA), increased urinary nitrate/nitrite, and decreased blood glutathione (GSH). Deferiprone (or L1), an orally active iron chelator or tetrahydrocurcumin (THU), a reduced derivative of curcumin from Curcuma Longa (turmeric), at dose of 50 mg/kg/day was administered intragastrically though the period of iron overload induction can alleviate these effects by restoring blood pressure, decreasing oxidative stress and enhancing antioxidant GSH significantly (p<.0.05 vs. iron overloaded mice). Moreover, combined therapy with L1 and THU exert a greater effect than L1 or THU monotherapy. Overall findings of this study suggest the beneficial effects of L1 and THU on reducing blood pressure, oxidative stress and also reveal the iron chelating capacity of THU in iron sucrose-induced iron overload in mice.
