Abstract Cholangiocarcinoma is an aggressive tumor of bile duct epithelium for which a standard effective treatment is unavailable. In this study, the cytotoxic effect of the crude ethyl acetate (EtOAc) and methanol (MeOH) extracts of Stephania venosa tubers in human CCA cell lines were investigated. The EtOAc extract exhibited cytotoxicity against KKU–M139, KKU–M213 and KKU–M214 cells with IC50 values of 15.68 ± 0.23, 11.79 ± 0.37 and 7.50 ± 0.42 μg /mL, respectively. The MeOH extract exhibited cytotoxicity against KKU-M139, KKU–M213 and KKU–M214 with IC50 values of 7.25 ± 0.34, 9.28 ± 1.0 and 0.61 ± 0.15 μg /mL, respectively. Chromatographic separation of these crude extracts yielded 5 compounds. Their structures were identified by spectroscopic mean to primary conclude as alkaloid type. These compounds thus, hold the promise of being a potent anticancer agent that deserved further exploration.
