     Abstract Berberine has been traditionally used for the treatment of diabetes mellitus. Many studies have shown hypoglycemic activity of berberine. However, its impact on sex hormone synthesis pathway is still less. The present study aims to investigate effect of berberine on the expression of sex hormone synthesis related genes in β-naphthoflavone (BNF)-exposed mouse testes. Adult male C57BL/6Mlac mice were consecutively intragastric administered berberine (7.5 mg/kg/d) for 7 day, and/or intraperitoneally injected BNF (30 mg/kg/d) for the last 3 days. Expression of testicular CYP17, CYP19, 3β-HSD1, 17β-HSD3, and 17β-HSD1 mRNAs were semi-quantified by RT-PCR. Berberine significantly up-regulated the expression of CYP17, 3β-HSD1, and 17β-HSD1 mRNA while BNF markedly induced the expression of 17β-HSD1 mRNA. Regarding to the BNF-exposure, berberine reduced 17β-HSD1 mRNA to nearly the same level as the normal whereas those of CYP17 and 3β-HSD1 were as high as that did by berberine itself. These observations suggested that berberine disturbed sex hormone synthesis pathway; consequently possibly resulted in modification of estrogen pattern in the body. Therefore, a caution is noted for the use of berberine as an alternative medicine and a further study to affirm the evidence in the DM status is worth.
