Abstract |
Introduction
The oxidant production is occurred when the homeostasis is disturbed, resulted in oxidative damage to tissues and multiple organs. Social isolation is a cause of oxidative stress. The harmful process can be indicated by increasing level of lipid peroxidation and decreasing of glutathione system. Curcumin is a major active constituent in rhizomes of Curcuma longa L. It has been widely used as a food additive and also an traditional herbal medicine throughout Asia. Therefore, the present work aims to investigate impact of curcumin on the oxidant-antioxidant system in the livers of mice with social isolation induced acute stress.
Experimental
Male ICR mice at 3 week of age were induced acute stress by social isolation (SI) for 6 weeks, and then the SI mice were divided into 4 groups (n=5) including the control group daily receiving 0.5% carboxymethylcellulose in 0.9% normal saline solution (CMC+NSS) and the treatments with curcumin (10 or 20 mg/kg/day), or imipramine (20 mg/kg/day), for 2 weeks. The normal mice (n=5) were daily given CMC+NSS for the same period. Twenty four hours after the last treatment, the livers were immediately collected and kept at-80°C for measuring the level of lipid peroxidation and the glutathione contents.
Results & Discussion
SI extensively elevated the lipid peroxidation in the mouse livers while curcumin at 20 mg/kg/day significantly recovered the level of MDA production in the SI mouse livers comparable to those levels of the normals. Correspondingly, SI excessively increased the oxidized glutathione content and reduced the ratio of reduced to oxidized glutathione (GSH/GSSG) in the livers of SI mice. Interestingly, curcumin significantly raised the GSH/GSSG ration in the livers of SI mice back to the same level as that in the normals.
Conclusion
These findings revealed that curcumin exerted antioxidation potential by decreasing the level of MDA production and increasing of the GSH/GSSG ratio in the livers of mouse with acute stress, leading to be a promising alternative candidate against SI induced oxidative stress.
