     Abstract This work studied the statistical optimization of dilute sulfuric acid hydrolysis (H2SO4) of two-stage pretreated cassava stem using Response Surface Methodology to obtain the hydrolysate containing high reducing sugar and low inhibitor concentrations. The optimum hydrolysis conditions that resulted in the highest total reducing sugar yield were H2SO4 concentration of 0.20 M, ratio of acid volume to sample of 16.71 mL/g and hydrolysis time for 90 min. These optimum conditions allowed obtaining a hydrolysate rich in reducing sugar, with a yields up to 67.37%. Conversely, inhibitors including furfural, 5-hydroxymethylfurfural, acetic acid and phenolic compounds were detected at very low concentrations.The fermentation of the hydrolysate resulted in an ethanol yield of 22.58 g/100 g substrate corresponding to a theoretical ethanol yield of 84.41%. The results demonstrate that the hydrolysate under optimum conditions containing highly fermentable sugar and low inhibitor concentrations is a promising source for high yield cellulosic ethanol production.
