Abstract |
Bacteria in the genus Bacillus have increasingly gained much interest as probiotics supplemented in human and animal diets. This is mainly due to their ability in producing spores that well survive the adverse environmental conditions. The aim of this work was to evaluate the medium compositions for cell and spore production of the probiotic Bacillus sp. B51f. The one-factor-at-a-time experiments were performed to examine the effects of inorganic nitrogen sources [(NH4Cl, (NH4)2SO4, NH4H2PO4, KNO3], carbon and energy source (cassava starch), growth factors (crushed brewer‘s yeast: CBY) and trace elements (MgSO47H2O, MnSO4H2O, CaCl22H2O) in supporting bacterial cell growth and sporulation of Bacillus B51f. The results showed that the medium containing 1% NH4Cl, 2% tapioca starch and 0.25% CBY yielded the highest growth of Bacillus B51f at a greater than 2x108 CFU ml-1 after 2 days of incubation. It was also found that the medium containing the combination of MgSO47H2O (0.05%), MnSO4H2O (0.005%) and CaCl22H2O (0.01%) significantly enhanced spore yield (107-108 spores ml-1) when compared to the commercial Nutrient broth (106 spores ml-1). Therefore, the optimal medium compositions for producing cells and spores of Bacillus sp. B51f were 1% NH4Cl, 2% tapioca starch, 0.25% CBY, 0.05% MgSO47H2O, 0.005% MnSO4H2O and 0.01% CaCl22H2O. |
