บทคัดย่อ |
Sweet sorghum stalks with the peel (6 to 20 mm in diameter and 6 mm in thickness) were used as low cost support materials for immobilization of Saccharomyces cerevisiae NP 01 to produce ethanol. To immobilize yeasts, the sterilized stalks were mixed with yeast extract malt extract (YM) medium containing yeast cell concentration of 1×108 cells ml-1 under static condition at 30 oC for 24 h before use as an inoculum for ethanol production. The ethanol production medium was sweet sorghum juice containing total sugar of 240 g l-1 without any nutrient supplement. Batch ethanol fermentation by the immobilized yeasts was carried out in 500-ml air-locked Erlenmeyer flasks under static condition at 30 oC. At the end of the fermentation, the cell concentration on the stalks increased from the beginning (3.76×108 cells g-1 dry weight of stalk) to 6.52×108 cells g-1 dry weight of stalk. The ethanol concentration (P), productivity (Qp) and yield (Yp/s) were 78.50±1.78 g l-1, 1.31±0.01 g l-1 h-1 and 0.45±0.01, respectively. Continuous ethanol fermentation by the immobilized yeasts on the sweet sorghum stalk was also carried out in a two tubular packed-bed bioreactor at the total dilution rate of 0.013 h-1. The results showed that the continuous system could be operated at least 432 h without any losses in the ethanol production efficiencies. The cell concentration in the broth remained constant at about 5×107 cells ml-1. After 432 h, the shape of the support materials was not different from the beginning and the average P, Qp and Yp/s were 100.34±1.86 g l-1, 1.30±0.02 g l-1 h-1 and 0.47±0.01, respectively. The results obtained clearly demonstrated that sweet sorghum stalk is suitable as a support material for yeast cell immobilization and can be applied in pilot and industrial scales for ethanol production. |
