| Abstract |
Alcoholism is one of the important risk factors of many diseases and deaths worldwide. Today, certain anticonvulsants are used in the treatment of alcohol withdrawal syndromes such as topiramate, sodium valproate and levetiracetam. Acorus calamus Linn. (AC) is an aromatic medicinal plant which has anticonvulsant effect. Base on the mechanism of action, AC might be use for treatment of alcohol withdrawal syndromes. Adult male Wistar rats were used in this study. Ethanol (7.2% v/v) was given to rats in a liquid diet, equivalent to ethanol 10.23 g/kg/day, at 10.00 a.m. every day for 16 days. On Day 17 at 9 a.m., animals were forced fed with either 3% Tween 20 (control), topiramate 50 mg/kg (positive control), AC 200 or 400 mg/kg. Every hour after alcohol withdrawal signs including stereotyped behavior, agitation, abnormal posture, abnormal gait andlocomotor hyperactivity were observed. The results showed that topiramate 50 mg/kg, AC 200 or 400 mg/kg significantly reduced stereotyped behaviors, agitation, abnormal posture and abnormal gait. Moreover, two doses of AC significant reduced locomotor hyperactivity including number of crossing, nose poking and rearing but topiramate increased locomotor activity by elevated number of crossing and rearing. The results suggested that β-asarones as a major compound of AC that enhancing tonic GABAergic inhibitionand attenuated the alcohol withdrawal symptoms. Thus, Acorus calamus might be one of medicinal plant for treatment alcoholism. |
