Abstract |
Streblus asper (SA) is one of the medicinal plants used in many countries which has anti-oxidative and anti-inflammatory properties. Previously, SA seed has been reported for acetylcholinesterase inhibitory activity. This study aimed to investigate the effects of SA leaf extract on brain acetylcholinesterase activity and memory in mice treated with scopolamine (SCO). Male ICR mice were divided into 6 experimental groups including Control, SA200, SA400, SCO, SCO+SA200 and SCO+SA400 groups. Animals were pretreated with SA 200 or 400 mg/kg/day by oral gavage once daily for 2 weeks. Memory deficits were induced by SCO (1 mg/kg, i.p.) 15 min before Morris water maze test. At the end of the test, mice’s brain was collected and acetylcholinesterase (AChE) enzyme activity was determined. The results showed that escape latency time in SCO group were significantly higher than control group. Interestingly, SA extract could significantly reverse scopolamine-induced memory deficits by reduce
escape latency time in Morris water maze. The activity of AChE enzyme in the brain of animals received SCO plus SA was significantly decrease when compared to the animals received SCO without SA. In conclusion, SA leaf extract can ameliorate scopolamine-induced memory deficits in Morris water maze test. In addition to anti-oxidative and anti-inflammatory properties as reported by the others, SA could possibly decrease AChE enzyme activity and augments cholinergic activity in the brain. These results suggest that SA leaf extract might be useful as an additional or alternative treatment in memory impairment. |
