| Abstract |
Pomelo (Citrus grandis L. Osbeck), the largest of all citrus fruits is the native fruit of great economic in Thailand. The study aimed to assess antidepressant effect of extract from juice and juice sac of Tubtim Siam (TS) pomelo on unpredictable chronic mild stress (UCMS). UCMS has been used as a model of depression in animal model. UCMS paradigm involves the exposure of mice to a variety of relatively mild unpredictable stressors in a random order for 6 weeks. Three weeks after stating the UCMS procedure, mice were administered TS daily (250 and 500 mg/kg, p.o.) or imipramine (20 mg/kg, i.p.). Antidepressant activity was investigated using tail suspension test (TST), forced swimming test (FST), locomotor activity test and level of serum corticosterone. The extract of TS of both doses significantly decreased immobility time in both TST and FST without the effect on locomotor activity as well as giving imipramine. TS extract also significantly reduced the level of serum corticosterone in mice. The results of this study suggest that TS extract seems to be mediated by decreasing the activity of hypothamic-pituitary-adrenal axis in hippocampus brain. |
