     Abstract Introduction: Recently, the antifertility studies of plant or herb are conducting to search for male contraceptive plants that can reduce the side effects from synthetic contraceptives. Although Leucaena leucocephala (LL) can cause hair loss and infertility in cattle, the antifertility effects of LL have never been systemically reported. Therefore, this study aimed to investigate effects of LL aqueous shoot tips plus young leaves extract on reproductive system in male rats. Methods: Shoot tips plus young leaves of LL were harvested in Khon Kaen province. Ten male rats were divided into two groups; control group was orally received with distilled water and LL group was received with 1500 mg/kg BW of LL extract for consecutive 60 days. Results and Discussion: The results showed that LL-treated rats had significant increases of malondialdehyde level, percentage of sperm acrosome reaction, and abnormal sperm head as compared with the control. Additionally, LL extract significantly reduced the sperm count, follicle-stimulating and testosterone hormone levels, and expression of testicular steroidogenic acute regulatory (StAR) protein. Moreover, seminiferous tubular atrophy and few germ cells in some tubules were observed in LL-treated rats. However, the extract had no adverse effect on body and reproductive organ weights, luteinizing hormone level, and expression of tyrosine phosphorylated proteins. Conclusion: These findings suggested that aqueous shoot tips plus young leaves extract of LL has male antifertility property in rats.
