     Abstract Introduction: Stress adversely affects male reproductive organs, causing infertility. Although various parts of Phyllanthus emblica L. (PE) extract have been shown to have high antioxidant capacity, protective effects of PE leaves extract on male reproductive parameters in chronic stress have never been elucidated. This study aimed to investigate effects of PE aqueous leaves extract on testicular protein markers in chronic stress rats. Methods: Male rats were divided into 4 groups (control, chronic stress [CS], PE 50, and 100 mg/kgBW+CS). In treatment group, animals were fed with different doses of PE extract before chronic stress induction by immobilization (4h/day) and forced-cold swimming (15 min/day) for 40 consecutive days. The sperm quality, testicular histology, and levels of corticosterone, testosterone, and malondialdehyde (MDA) were examined. The expressions of testicular steroidgenic acute regulatory (StAR) and tyrosine phosphorylated proteins were analyzed by immuno Western blot. Results: PE significantly increased sperm concentration and decreased corticosterone and MDA levels, sperm head abnormality (%), and acrosome reacted sperm (%) in CS rats. PE could also minimize testicular histopathology in CS rats. Moreover, PE altered the intensity of tyrosine phosphorylated proteins (14, 51, and 60 kDas), except of StAR protein in CS testis. Conclusion: PE leaves extract can prevent testicular damages in chronic stress rats.
