     บทคัดย่อ Metabolic syndrome (MetS) is a group of metabolic risk factors for cardiovascular diseases (CVD). Oxidative stress is a common pathophysiological process related to MetS. Glutathione peroxidase-3 (GPX3) is an extracellular antioxidant which plays role in the detoxifications of hydrogen peroxide. Single-nucleotide polymorphisms (SNPs) in GPX3 gene, rs1946234 and rs3828599, have been reported that affect gene expression which may contribute to the development of MetS. The objective of this study was to assess the associations of these GPX3 polymorphisms with MetS in community-based subjects residing in Khon Kaen. A total of 454 subjects (mean age 51.0±9.9 years, 207 males and 247 females) were recruited. The prevalence of MetS was determined according to modified National Cholesterol Education Program III criteria. SNPs were genotyped using allele specific polymerase chain reaction (AS-PCR). The prevalence of MetS was significantly higher in females (67.3%) than in males (32.7%). Both SNPs revealed association with the risk of MetS; the respective OR(95%CI) for rs1946234-C allele carriers and for individuals with rs3828599-GG were 1.60 (1.01-2.53) and 2.14 (1.14-4.02). The combination of rs1946234-(AC+CC) with rs3828599-GG further increased the risk of MetS with OR(95%CI) = 3.80 (1.73-8.37). The present study demonstrated the associations of GPX3 polymorphisms with the risk of MetS in Khon Kaen community subjects. These might result from the low level of GPX3 expression with less protective capacity against oxidative stress in subjects with the GPX3 risk alleles.
