     Abstract An association between oral health diseases and hypertension has been proposed. It has been reported that an administration of L-arginine analogues, NG-nitro- L-arginine methyl ester hydrochloride (L-NAME), a substance widely used in animal hypertensive model could decrease salivary flow rate and total protein concentration. However, little information regarding the other biochemical molecules in salivary gland under hypertensive condition has been reported. Therefore, this study we aimed to detect the effect of hypertension on rat salivary glands using different four methods; conventional microscopic technique, matrix-assisted laser desorption/ionization (MALDI-TOF), liquid chromatography-mass spectrometry (LC-MS) and Fourier transform infrared (FTIR) micro spectroscopic technique. Male Sprague-Dawley rats were given 40 mg/kg/day of L-NAME in drinking water to induce hypertension for three weeks. The submandibular and sublingual glands from normal and hypertensive rats were used in this study. Our results showed that histological structure of submandibular and sublingual glands has non-significant difference between control and hypertensive group. However, FTIR showed that salivary glands in the hypertensive group have higher concentrations of triglycerides, carbohydrates and proteins than those in control group. In addition, MALDI-TOF technique evidently discreted the salivary glands protein profiles between two groups. Interestingly, the expression of phosphoproteins using LC-MS showed only one identified protein in control group of submandibular gland and 6 proteins of sublingual glands whereas 7 identified proteins were found in hypertensive group of submandibular gland and 8 proteins of sublingual glands. The finding of this study could provide the preferred method for initial detection the abnormal of salivary gland associated with hypertension.
