     Abstract Mutation of the thermotolerant yeast Saccharomyces cerevisiae DBKKUY-53 was induced using ethyl methane sulfonate (EMS) and its mutant strains that capable of growth at high sugar concentrations were screened using yeast extract malt extract (YM) medium containing different glucose concentrations. S. cerevisiae wild-type strain (DBKKUY-53) exhibited a good growth in the YM medium containing 250 and 300 g/L glucose. When sugar concentration in the YM medium increased to 350 and 400 g/L, growth of the yeast strain was reduced. By using EMS mutagenization, 42 isolates of the S. cerevisiae mutant strains were obtained. Comparative study on growth performance at high temperature (37C) of the selected mutant and wild-type strains using YM medium containing 350 g/L glucose was performed. As found in the current study, a mutant strain designated S. cerevisiae EMS33 exhibited a good growth under a selective condition. Morphological analysis and growth properties of the selected mutant strains were performed and described in this work.
