Khon Kaen University, Khon-Kaen, 40002, Thailand Introduction: Rice bran contains natural phytochemicals which has been reported to possess antioxidant, anti-inflammatory, anti-hypertension, improve insulin resistant and anticancer. Rice bran hydrolysates (RBH) were prepared from proteolysis of rice bran to improve functional properties. RBH have been suggested to have immunomodulatory effect, however, the effect is not well established. The present study aimed to explored effects of RBH from Tubtim-Chum-Phae rice bran on T-lymphocytes. Materials and Methods: Jurkat cells, the T lymphocytes, were used to evaluate RBH immunomodulatory properties. The cells were cultured in RPMI 1640 supplemented with 10% fetal bovine serum and antibiotics. Cells were cultured with RBH at varied concentrations for 72 h before cell proliferation was assessed by MTT assay. The effect on chemotaxis was assessed by transwell chamber assay. The cells were incubated with RBH in serum-free medium on the insert chamber coated with Matrigel, while the lower chamber contained with full medium. The number of Jurkat cells migrated through the insert was assessed. Protein from cell lysates of Jurkat cells were prepared after treatments and subjected to Western immunoblotting. Results: RBH showed antiproliferative effect on T-lymphocytes in a dose-dependent manner. RBH suppressed chemotaxis of lymphocytes by inhibition of cell migration in transwell assay. The antiproliferative and anti-chemotaxic activities were associated with activation of AMPK and suppression of the protein expression of mTOR and PI3K. Conclusions: RBH showed the immunomodulatory effect on T-lymphocytes by demonstrating antiproliferation and suppression of chemotaxis. Consumption of rice bran may provide health benefit on alleviation of allergic reaction.
