| บทคัดย่อ |
Gold nanoparticles (AuNPs) is growing up to use in biomedicine due to inert property making AuNPs susceptible to react with the cellular metabolism resulting to the particle degradation and low toxicity in tissues. The understanding of physicochemical characteristics of AuNPs influence its interaction with biological systems is required, particularly in innate immune cells. This study investigated the effect of different surface charges of AuNPs with same size and shape on internalization mechanism by using ICP-MS and cell activation by using western blot and ELISA. In the results, the different surface charges AuNPs were differently responses including the pathways of internalization, cell activation and inflammation. The positive charged AuNPs (AuNPs+ve) tended to be taken up via various pathways including macropinocytosis, clathrin-mediated endocytosis and caveolae-mediated endocytosis, whereas the negative charged AuNPs (AuNPs-ve) might be internalized through macropinocytosis and clathrin-mediated endocytosis. Interestingly, the AuNPs+ve have strongly provoked the cellular response to produce IL-6 compared with AuNPs-ve stimulation. These results might be importance data to design nanoparticles to target or avoid the innate immune system in nanomedicine in future. |
