     Abstract Dengue nonstructural 1 (NS1) is one of an attractive candidate antigen to be incorporated in dengue vaccine due to its ability to elicit protective immunity that can cross-react with all types of dengue virus. In this study, the codon-optimized NS1 gene was cloned in our Escherichia coli-L.casei shuttle vector using a constitutive promoter of lactate dehydrogenase gene, pRCEID-LC13.9. The two recombinant plasmid containing NS1 gene, pRCEID-LC13.9:NS1 without signal peptide and pRCEID-LC13.9:USP:NS1 carried the signal peptide, Usp45, were constructed and were each transformed in the expression host, L. casei RCEID02. It was found that the recombinant L. casei carrying pRCEID-LC13.9:NS1 and that carrying pRCEID-LC13.9:USP:NS1 can successfully expressed NS1 protein as intracellular and secreted form as determined by immunoblotting using anti-NS1 antibody.
