Research Title A Mixture of Palmitic Acid and Oleic Acid Potentiates the Expressions of Lipid Metabolism Enzyme CYP4A11, Peroxisome Proliferator-Activated Receptor Alpha, and Hepatic Drug Transporter SLCO1B1 mRNA in HepG2 cells

     Abstract A high fat-diet is one of the important factors in progression of chronic diseases such as obesity, dyslipidemia and non-alcoholic fatty liver disease (NAFLD). Palmitic acid (PA) and oleic acid (OA) are fatty acids mostly found in daily dietary. Liver is the main organ that plays an important role on lipid metabolism and biotransformation of antilipidemic drugs. This study aims to examine the effects of PA and OA on the mRNAs expression of a lipid metabolizing enzyme CYP4A11, peroxisome proliferator-activated receptor alpha (PPAR), and a hepatic drug transporter protein (SLCO1B1 gene/OATP1B1) in hepatocellular carcinoma (HepG2). HepG2 cells were cultured in a 6-well plate (5x105 cells/well) and treated with 1 mM PA and/or OA in 0.2% isopropanol for 24 hours. Expressions of CYP4A11, PPAR, and SLCO1B1 mRNA were determined by RT-qPCR. Either PA or OA did not alter the expression of CYP4A11, PPAR, and SLCO1B1 mRNA while the mixture of PA and OA (1:1) significantly increased the mRNAs expression of all; CYP4A11 and PPAR at p<0.05 and SLCO1B1 at p<0.001. Hence, excessive intake of high fat-diet containing both saturated and unsaturated fatty acids might induce the expressions of the genes related to lipid metabolism and biotransformation of clinical drugs in human liver, subsequently resulted in a risk of drug interaction and undesirable clinical outcomes.
