     Abstract Spent yeast from bioethanol fermentation process is an important source of beta-glucans which are linear chain of glucose residues polymerized through β-1,3 β-1,4 or β-1,6 glycosidic linkages. Among several benefits on biological response, a function as immunostimulants to protect cells against infectious disease is from beta-1,3-glucan. In this study, the β-glucan was extracted from Saccharomyces cerevisiae TISTR 5339 biomass and subjected to the Fourier transform infrared (FTIR) spectroscopy for composition analysis. The result showed that the polysaccharide content was 64.9% from autolysis. FTIR spectrum indicated that the content of beta-1,3-glucan (~1048 cm-1, wavenumber) was 39.6 % and beta-1,6-glucan (~998cm-1, wavenumber) was 33.8% of total polysaccharide. Therefore, FTIR could be applied for studying the structure of beta-glucan and confirmed that S. cerevisiae TISTR 5339 spent yeast composed of high beta-glucan content with the ratio of beta- 1,3-glucan per beta-1,6-glucan of 1.17.
