Metformin prevents memory deficits and reductions in cell proliferation induced by methotrexate chemotherapy in adult rats |

บทคัดย่อ |
Metformin (1,1-dimethylbiguanide hydrochloride) is currently used as the first-line drug to treat patients with type 2 diabetes. Previous studies have demonstrated that metformin can reduce neuroinflammation and hippocampal neuronal cell loss, which can subsequently improve spatial memory. Methotrexate (MTX) is one of chemotherapeutic agents in the antimetabolite group. It is commonly used to treat various types of cancer, which is associated with cognitive impairment in many patients. Therefore, the aim of the present study was to investigate the preventive effects of metformin on a reduction of memory and hippocampal cell proliferation induced by MTX in a rat model. Male Sprague-Dawley rats were divided into 5 groups, including control, MTX, metformin (Met), preventive, and throughout groups. MTX (75 mg/kg BW) was administered by intravenous
injection on days 7 and 14. Rats were administered with Met (200 mg/kg BW) by intraperitoneal injection for 14 days. Some of the MTX-treated rats received co-treatment with Met once a day for 14 days (preventive group) and 28 days (throughout group). After the drug administration, memory was determined using novel object location (NOL) and
novel object recognition (NOR) tests. Furthermore, cell proliferation in the subgranular zone (SGZ) of the dentate gyrus in hippocampus was quantified by Ki-67 immunostaining. The results showed that MTX chemotherapy impaired memory and reduced cell proliferation in the SGZ of the hippocampal dentate gyrus. Rats in Met, preventive and throughout groups showed significant memory impairments and an increases of Ki-67 positive cells. These data demonstrate that metformin can prevent memory and hippocampal neurogenesis impairments caused by MTX chemotherapy. |
