     Abstract Dehydroepiandrosterone sulfate ;DHEASͿ is the most abundant circulating steroid hormones which plays an important role as the initial substance for steroid hormones synthesis͘ The measurement of the DHEAS level is used as an ͚ agingͲ biomarker͛ to assess the risk of diseases associating with age͘ In general, the enzymeͲ linked immunosorbent assay ;ELISAͿ is most widely used for DHEAS measurement because of highly specific, rapid, and suitable detection͘ But the commercial ELISA kit for DHEAS detection has not been produced in Thailand͘ So, expenses of using imported commercial ELISA kit were quite high and not suitable in resourceͲlimited laboratories͘ Therefore, this study aimed to develop and standardize a costͲ effective inͲ house competitive ELISA for DHEAS detection͘ An inͲhouse competitive ELISA format was developed and optimized using a polyclonal antiͲDHEAS as the antibody coating͘ DHEASͲHRP conjugate and 3,3഻,5,5഻Ͳtetramethylbenzidine ;TMBͿ were used as a competitor and the enzyme mediatorͬsubstrate system, respectively͘ Under optimal conditions, the standard curve was established from standard DHEAS concentration range of 100 Ͳ 10000 ngͬmL which a linear regression and the limit of detection was 97͘947 ngͬmL͘ The result covers normal blood levels of DHEAS since the fetus to the age of 59 years old ;120Ͳ 3210 ngͬmLͿ͘ Furthermore, the inͲhouse competitive ELISA was estimated to be 3 times less per test compared to the cost of imported commercially available ELISA kit͘
