     Abstract Chlorella sp. biomass was pretreated by acid-hydrothermal method. Optimum conditions for the pretreatment were 1.5% (v/v) HCl, temperature of 180 °C, and reaction time of 15 min. Under these conditions, a maximum reducing sugar yield of 162.8 mg/g-dry-biomass was attained. The hydrolysates were further used as a sole substrate for bio-hydrogen production by dark fermentation. A maximum bio-hydrogen yield of 47.2 mL/g-volatile-solids (VS) was achieved at the initial pH of 6.0, and feedstock to inoculum ratio of 1.0 on a VS basis. Results indicated that Chlorella sp. biomass is a feasible feedstock for bio-hydrogen production, and that acid-hydrothermal pretreatment is effective in enhancing bio-hydrogen production from the biomass.
