Abstract |
The objectives of this study were to investigate whether lipopolysaccharide of P. gingivalis (LPS-Pg) could induce autophagy in human gingival fibroblasts (HGFs) and to analyze whether Vicenin-2 (VCN-2), a flavonoid glycoside, exhibited any effect on autophagy induced by LPS-Pg in HGFs. The PrestoBlue assay demonstrated that different concentrations of LPS-Pg (1.5-100 µg/mL) and VCN-2 (1.5-400 µM) did not significantly affect the cell viability of HGFs. Among all concentrations tested, 5 µg/mL of LPS-Pg showed the highest induction of autophagy in HGFs, confirmed by an increase in the protein expression levels of LC3B-II/LC3B-I ratio and a reduction of p62 protein levels. The immunofluorescent study demonstrated that VCN-2 at concentrations of 12.5 and 25 μM can increase the levels of p62 protein in LPS-Pg-treated HGFs. These results preliminarily provide evidence of VCN-2’s ability to inhibit the induction of autophagy induced by LPS-Pg, suggesting the potential use of VCN-2 to be a novel alternative modality for periodontal disease treatment. |
