     Abstract Objective: To study the population genetic of cat- and human-specific Opisthorchis viverrini (O. viverrini) in cyprinid fish in Khammouane, Lao PDR and Udon Thani, Thailand. Materials and Methods Cyprinid sp. fish were collected from Khammouane, Lao PDR and Udon Thani, Thailand from July 2022 to February 2023. Fish were digested using homogenization and an acid-pepsin digestion procedure. Finally, a stereomicroscope was used to isolate and identify the metacercariae in the silt. O. viverrini-like metacercariae were isolated individually and added 1X PBS in 1.5 ml microcentrifuge tube. Individual O. viverrini-like metacercaria was extracted and positive O. viverrini metacercariae were sent for sequencing. Cytochrome c oxidase subunit 1 (cox1) was used to investigate genetic variation in the present study. Results: The sequencing results; The phylogenetic tree analysis of 91 and 53 individual O. viverrini metacercariae from Khammouane, Lao PDR and Udon Thani, Thailand, respectively revealed that all O. viverrini metacercariae in cyprinid fish belonged to human-specific O. viverrini. Cat-specific O. viverrini could not be found in cyprinid fish from Udon Thani and Khammouane. We got 99.25 to 100% identity with humanspecific or widespread cluster O. viverrini from BLASTN analysis. The results were also supported by the median-joining haplotype network analysis. Conclusion Our results revealed that all O. viverrini metacercariae from cyprinid fish in Khammouane, Lao PDR and Udon Thani, Thailand, belonged to human-specific O. viverrini. None of our samples belonged to cat-specific O. viverrini. We assumed that the cox1 nucleotide sequence from cat-specific O. viverrini metacercariae which is identical to cat-specific O. viverrini eggs cannot be found in the environment, due to small amounts of sample size or possibly harbored in the different secondary intermediate hosts. In our opinion, cyprinid fish seems to be accidental host of cat-specific O. viverrini metacercariae. Further investigation is needed to answer the question; what and where is the main secondary intermediate host of cat-specific O. viverrini?.
