Abstract |
Background/Aim(s): Ethanol withdrawal after prolonged ethanol consumption increases anxiety-like behaviors in rodents which might reflect ethanol dependence in humans. Mucuna pruriens (MP) has long been used in traditional medicine for various neurological diseases as it has antioxidant, neuroprotective, and neurorestorative effects that help promote brain health. In the present study, we investigated the effects of Thai Mucuna pruriens (T-MP) seed extract on anxiety-like
behaviors following ethanol withdrawal in mice.
Methods: Mice were exposed to ethanol (2g/kg/day of ethanol, i.p.) for 10 consecutive days. Mice were tested for anxiety-like behavior in light/dark box (LDB) and elevated plus maze (EPM) tests at 12, 24, and 36 h after the last dose of ethanol. At the maximum effect-time point, 1 h before the tests, T-MP seed extract (600 mg/kg, p.o.) or diazepam (4 mg/kg, i.p., as a positive control) was given to the animals.
Results: In both LDB and EPM tests, the anxiety-like effect of ethanol withdrawal was clearly seen at all times tested with the maximum effect at 24 h. As a result, the effects of T-MP seed extract and diazepam, given 1 h before the 24-hour point were examined. The results showed that both TMP and diazepam could significantly reverse the anxiety-like effect of ethanol withdrawal in either LDB and EPM tests in a comparable manner.
Conclusion: Results of this study suggest that T-MP (600 mg/kg) might be effective in managing the anxiety-like effects of ethanol withdrawal. MP seeds contains many active substances including levodopa, coenzyme Q10, and nicotine adenine dinucleotide. It is interesting to further investigate the relationship among the substances and the effects observed. |
