     Abstract Polycystic ovary syndrome (PCOS) is one of the common female endocrine disorders that lead to infertility. One of the known mechanisms of PCOS is the abnormally high level of androgen relative to estrogen. Androgen transduces its adverse e#ects in PCOS females through the androgen receptor (AR), the level of which increases accordingly. A decrease in AR is therefore desirable in an e#ective PCOS therapeutic treatment. !ai Mucuna pruriens (T-MP) seed extract is a medicinal plant previously used in infertility treatment. We have recently shown that it increases estrogen in healthy female rats. !erefore, we hypothesized whether T-MP could reverse abnormal symptoms in letrozole-induced PCOS rats. !is study was approved by the National Research Council of !ailand (No. IACUC-KKU-54/65). Normally cycling female rats (n=28) were treated with letrozole (1mg/kgBW daily, p.o.) for 21 days. For the following 30 days, 21 letrozoletreated rats were further p.o. administered with both letrozole (same dose as before) and T-MP (150, 300, or 600 mg/kgBW) (n=7 in each group). Another 7 rats with no letrozole/ T-MP treatment served as controls. Polycystic ovaries were histologically observed in rats 21-day-treated with letrozole (Let) with signi"cant increases in body weight and percentage of relative weight in ovary but a decrease in uterine, compared with controls, and their AR level increased to 1.44X the control value. Let-150-mg-T-MP treated rats showed the same histologically de"ned PCOS and ovary weight as the Let rats and their AR level rose to 1.63X the control value. However, ovarian cystic follicles were much decreased in Let-300-mg-T-MP rats and became zero in Let-600-mg-T-MP. Notably, in Let-600-mg-T-MP rats, the body and ovary weight were not signi"cantly di#erent from controls and the AR level was only 1.22X the control value. Our results suggest that T-MP might be an alternative agent to be further studied for PCOS treatment.
