Research Title Characterization of a fragrant gene and enzymatic activity of Betaine Aldehyde Dehydrogenase in aromatic and nonaromatic Thai rice cultivars

     Abstract Betaine aldehyde dehydrogenase 2 (Badh2) is known to be a fragrant rice gene. An 8-bp deletion and three single nucleotide polymorphisms (SNPs) in its exon 7 of the Badh2 gene are reported to influence fragrant level of rice (Bradbury et al., 2005). Nevertheless, only one Thai fragrant rice cultivar, KDML105, has been characterized for this gene. The 8-bp deletion also leads to malfunction of the BADH2 protein. However, there was also no report on rice betaine aldehyde dehydrogenase activity previously. Therefore, the objectives of this study were (1) to characterize the Badh2 gene in fragrant and non-fragrant of Thai rice cultivars and (2) to determine betaine aldehyde dehydrogenase activity. The results showed that the presence of Badh2 gene encoding betaine aldehyde dehydrogenase (BADH2) of the Thai rice cultivars revealed significant sequence polymorphism containing a total of 3 SNPs and 8 bp deletion in fragrant rice; KDML105 and RD6, whereas, non-fragrant rice; Niaw san-pah tawng and Sew-mae-jan did not have. Determination of betaine aldehyde dehydrogenase activity using 4-aminobutyraldehyde (AB-ald) and 3-aminopropionaldehyde (AP-ald) as substrates showed lower Vmax/Km in the fragrant rice cultivars than that of the non-fragrant rice cultivars, suggesting that aldehyde dehydrogenase might play important role in aroma synthesis in rice.
