Abstract |
Utilization of antioxidants from plant extracts in cosmetic products has been overwhelmed and might cause undesired effects with little or no evidence on permeation and toxicity. Flowers of damask roses (Rosa damascena), a sources of powerful antioxidants used as cosmeceuticals to protect aging of the skin, were extracted by hydroglycol. The extract was investigated on skin permeation using an in vitromodel by porcine skin and skin cytotoxicity by human epidermoid carcinoma cells (A431). Quantification of the permeant as total phenolic content by Folin-Ciocaleu assay indicated a permeation rate of 10.3 ± 5.4µg gallic acid equivalent (GAE)×cm-2×h-1 or 0.5 %×cm-2×h-1. Cell viability of A431 after 24h exposure to the extract by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromideassay gave IC50 of 0.81mgGAE×ml-1. Real – time monitoring by xCELLigence system indicated a dose – dependent cytotoxic manner. Interleukin 1-α, monitored after 15 min of cell exposure to the extract for skin irritation and measured by ELISA, was found to be 11.2 ± 1.5 pg/ml. The exposure dose which causes IC50 tends to be lower than that potentially irritated the skin after immediate contact. It leads to conclude that the extract of damask rose could permeate the skin and cause irritation or cytotoxic at dose – and time – dependent manner.
