     Abstract Zinc supplement in commercially-available health care products are numerous. Because of its antioxidant activity, zinc could regulate immune system, promote wound healing and act as a skin anti-inflammatory agent. However, it could also known as a toxic substance particularly when over-consumption is involved. This study aims to assess cytotoxicity of Zn2+ using a model of human forehead fibroblasts (HFF) at concentration up to as high as 200 mg/ml. As zinc is a protein binding substance, cell culture for protein bound model and MTT assay at 540 nm was used. The result showed that cell viability at all concentrations of zinc was higher than 75% and not significantly different (p > 0.05) when compared with negative control group. Morphological changes were observed at concentration of 1 mg/ml and higher. In conclusion, Zn2+ has no potential cytotoxicity to HFF but might cause impaired cell function and morphology.
