Objective of this study was to validate optimal mobile phase in Thin Layer Chromatography analysis of sex steroid hormone profiles in plasma of swamp eel, Monopterus albus. Investigations were divided into 2 steps; the first step was to study an optimization of mobile phase using 5 solvent mixtures includes 1) chloroform:acetone, 9:1 (v/v), 2) benzene:ethyl acetate:acetone, 6:1:1 (v/v), 3) chloroform: benzene:methanol, 10:3:1 (v/v), 4) benzene:ethyl acetate, 2:1 (v/v) and 5) chloroform: methanol, 10:1 (v/v). The second step was evaluation of plasma sex steroid profiles in M. albus, divided into three groups according to weight and total length of fish; as <100 g/ < 40 cm, 100 – 200 g/ 40 – 50 cm and > 300 g/ > 60 cm, respectively. The best of optimal mobile phase was chloroform:benzene:methanol. Plasma sex steroids detected in presumably female M. albus include progesterone, 17α-hydroxyprogesterone, testosterone, 17β-estradiol and unknown. However, similar approach did not detect plasma sex steroid profiles in presumably male. |
