     Abstract Endogenous inflammatory lipid mediators have been reported to play a central role in inflammation and implicated in cancer development. Among them, the pro-inflammatory lipid mediators, especially leukotrienes (LT) which are produced from arachidonic acid by 5-lipoxygenase (5-LOX), play a central role in cancer development and progression. Here, we aimed to study the expression of 5-LOX during cholangiocarcinogenesis using immunohistochemistry staining in the liver fluke infection-induced hamster CCA model. Additionally, the expression of 5-LOX in human CCA tissues was also investigated. The results shown that the percentage of 5-LOX positive inflammatory cells were increased in CCA-induced hamster group when compared with the untreated hamster group. Moreover, praziquantel (antiparasitic drug) treatment could decrease 5-LOX expression in CCA-induced hamster. Interestingly, high 5-LOX expression was correlated with the recurrence status of CCA patient. Therefore, the 5-LOX may play an important role in the CCA development and progression. Hence, the inhibition of 5-LOX expression may be served as a new strategy for CCA treatment.
