บทคัดย่อ |
Introduction: Morus alba L (Moraceae) leaves have been known for its hypoglycemic activities. Since the biosynthetic pathway of secondary metabolites in cell culture might differ from that of nature plant, therefore the cell suspension culture extracts from M. alba was investigated for activities related to glucose lowering properties in vitro.
Materials and Methods: MeOH extract from M. alba cell suspension derived from leaves and its fractions from silica gel column chromatography were investigated for alpha-glucosidase inhibitory effect and the ability of glucose uptake stimulation into L6 myotubes.
Results and Discussion: Crude extract did not stimulate glucose uptake into L6 myotubes, but inhibited the function of alpha-glucosidase IC50 17.83 µg/mL. Fractions from silica gel column which contained beta-sitosterol showed the highest glucose uptake stimulation of 56 0 at concentration of 200 µg/mL 500 nM insulin gave 53.3 stimulation. Fractions which consisted of resveratrol and some fractions with unknown compounds were found to possess stronger alpha glucosidase inhibitors than crude extract with IC50 4.7-8.0 µg/mL IC50 of acarbose 35.2 µg/mL. The loss of glucose uptake activities of crude extract from cell suspension might be from the different chemical compositions pattern between leaves and cell culture.
Conclusions: Fractionation of crude extracts could be successively removed the inactive compounds and obtained more potent hypoglycemic fractions. The active fractions which contained beta-sitosterol and resveratrol were along with their hypoglycemic activities of glucose uptake stimulation and alpha-glucosidase inhibition, respectively which have been previously reported.
