     Abstract Background and objective: Stress affects the female reproductive system and is a cause of anovulation leading to infertility. Although tyrosine phosphorylated (TyrPho) proteins have well been investigated in male reproductive organs and are associated with fertility, the expressions of TyrPho proteins in female reproductive organs such as ovary, oviduct, and uterus have never been demonstrated under normal or stress condition. This study aimed to localize and investigate the expression of TyrPho proteins in female reproductive organs of chronic stress rats. Methods: Female Sprague-Dawley rats were divided into control and chronic stress (CS) groups. The rats in the CS group were induced to be stress by immobilization (4 h/ day) and individually forced to swim on cold water (15 min/ day) for 60 consecutive days. At the end of experiment, the histological sections of ovary, oviduct, and uterus were observed and compared between groups. All organs were also used to be localized for the presence of TyrPho proteins by immunohistochemistry. In addition, the expression of such proteins in reproductive tissues was investigated and quantified by using immuno-Western blotting. Results: It was shown that weights of ovary and oviduct between groups were significantly different but that of uterus in CS group was significantly greater than control group. Histologically, the CS ovary showed damages of secondary follicles compared to those of control while the structural changes of CS oviduct and uterus were slightly observed. . In addition, the TyrPho proteins were localized in the tissues of ovary, oviduct, and uterus. immunohistochemical analysis showed that positive reactivity of TyrPho proteins expression in. Moreover, many TyrPho proteins (ovary = 72, 43, 28 kDas; oviduct = 170, 55, 43 kDas; uterus = 55, 43 kDas, respectively) were separated and identified. Interestingly, the expressions of all TyrPho proteins in oviduct and uterus and of 43 and 28 kDas in ovary were significantly increased in cs group. Conclusions: This study has demonstrated the presence of TyrPho proteins in female reproductive organs with increased expressions under stress condition. Such uncharacterized proteins investigated in this study might be involved in female infertility especially in chronic women.
