| บทคัดย่อ |
Forkhead box M1 (FOXM1) is the transcriptional factor related to cancer hallmarks. There are 3 FOXM1 isoforms; FOXM1a, 1b, and 1c. FOXM1b and 1c are the transcriptional active, whereas FOXM1a is inactive. The expression of FOXM1 isoforms and the impact in cholangiocarcinoma (CCA), however, remains unclear. In this study, the FOXM1 isoforms in patients’ CCA tissues and cell lines were investigated. The expression of FOXM1 isoforms was determined using conventional PCR in 40 CCA tumors and 13 para-tumor tissues. It was found that FOXM1a was commonly detected in all tumor and para-tumor tissues. In contrast, FOXM1b (25%) and FOXM1c (70%) were differentially expressed in tumors, and was absent in para-tumors. Using real-time PCR technique, the 3 isoforms were found with FOXM1c being the predominant in 5 CCA cell lines tested. The expression of FOXM1 isoforms in tumor tissues did not associated with clinical features of patients, however, the survival of patients with positive FOXM1c had significantly shorter survivals than those with negative FOXM1c with hazard ratio 2.57. In conclusion, FOXM1c can be a worse prognosis marker for CCA. |
