     บทคัดย่อ Melioidosis is classically characterized by pneumonia and multiple abscesses, with a mortality rate of up to 40%. This tropical disease caused by ingestion, percutaneous inoculation or inhalation of the Gram-negative soil saprophyte Burkholderia pseudomallei. The biofilm formation of the B. pseudomallei is influenced by environmental factors and may enhance the pathogenesis of the bacteria. To investigate the role of B. pseudomallei biofilm during pathogenesis, B. pseudomallei H777 (biofilm wild type) and M10 (biofilm mutant) were co-cultured with the human lung epithelial cells (A549) at MOI 10 at 37 o C for 1, 4, 8 and 12 h to determine the role of biofilm in bacterial adhesion, internalization and intracellular multiplication using antibiotic protection assay. The B. pseudomallei biofilm mutant demonstrated a significant impairment of A549 cell adhesion compared to the wild type (p < 0.05). These results suggest that the B. pseudomallei biofilm play an important role during the first step of bacterial pathogenesis to adhere to the host cells. However, the biofilm did not show any responsibility during bacterial internalization or intracellular multiplication in human lung epithelial cells (p > 0.05). (abstract only)
