     บทคัดย่อ [Background] Our previous study has shown that some macrophages in the tumor stromal of oral squamous cell carcinoma (OSCC) tissue expressed the immunosuppressive protein indoleamine 2, 3-dioxygenase (IDO) more strongly than did tumor cells. In addition, IDO can be secreted from EBV infected B cells and inhibit NK cell cytotoxicity. EBV components in infected cells as well as both of Epstein-Barr Virus (EBV)-encoded small RNAs (EBER); EBER-1 and EBER-2 can be also carried via exosomes which modulate biological changes in neighboring cells. [Materials and Methods] This study aimed to investigate the effect of EBERs-containing exosomes in epithelial cells on IDO induction. Two EBV positive cell lines (B95-8, P3HR1) were used for exosome isolation by using differential ultracentrifugation. qRT-PCR was used to determine the expression of EBERs, IL-6, and IDO. HEK293T cell line was used as recipient epithelial cells. EBERs-transfected HEK293T cell lines were used to confirm the effect of EBERs on IDO expression. [Results] Both EBER-1 and EBER-2 were detected in the culture supernatants and also in the purified exosome fractions of all EBV positive cell lines. EBV-modified exosomes induced an expression of both IL-6 and IDO in HEK293T cell after incubation with exosomes. Interestingly, the level IL-6 and IDO were further increased in a time and a dose-dependent manner in both of EBER-1 and EBER-2 transfected HEK293T cell. [Discussion] These results reveal that EBERs are secreted from EBV-infected cells via exosomes, internalized in the recipient epithelial cells and induces IDO expression through IL-6 pathway. This study suggest that exosomes from neighboring cells containing EBERs may promote the immunosuppressive microenvironment and induce oral cancer development.
