     Abstract The Northeast of Thailand has the highest incidence of cholangiocarcinoma (CCA), a bile duct cancer, worldwide. CCA in this region is associated with Opisthorchis viverrini infection. CCA patients usually present with late-stage disease. Current treatments for CCA give unsatisfactory results. New and more effective forms of therapy for CCA are urgently needed. Ursolic acid (UA), a natural triterpene, exhibits the anti-cancer activities, e.g., suppressing proliferation and inducing apoptosis. The objectives of the present study were to: i) evaluate the cytotoxic effect of UA against CCA cells; ii) investigate UA-induced biomolecular changes by synchrotron radiation-based Fourier-transform infrared microspectroscopy (SR-FTIRM); and iii) detect apoptosis using flow cytometry. The results showed that UA inhibited CCA cell proliferation in a dose- and time-dependent manner. The SR-FTIRM results revealed significant modifications in the absorption bands of proteins and lipids, supporting the ability of UA to induce apoptosis. Flow cytometric data confirmed apoptosis-inducing property of UA and membrane changes during apoptosis. Thus, UA may be a promising candidate for CCA treatment.
