     Abstract Introduction: Phenol red is a pH indicator to detect a change of culture medium from neutral to acidic condition but how phenol red influences the expression of cytochrome P450 enzymes (CYP) in a cell culture is limited. Hepatocarcinoma (HepG2) and adenocarcinoma (Caco-2) cells are widely used as the representative models for human liver and intestine, the major organs of drug metabolism particularly for oral drugs. Objective: To examine effects of phenol red in the culture media on profiles of CYP1A2, CYP2E1, CYP3A4, and CYP3A5 mRNAs in HepG2 and Caco-2 cells induced by rifampicin. Materials and methods: HepG2 (1.25×105 cells/well) and Caco-2 (5×104 cells/well) were cultured in a 24-well plate for 72 h. Then the cells were treated with 5 M rifampicin in phenol red or non-phenol red contained culture media for 72 h. The expressions of CYP1A2, CYP2E1, CYP3A4, and CYP3A5 mRNAs were quantitatively determined by RT/qPCR. Results: Rifampicin significantly induced the expressions of CYP3A4 and CYP3A5 in HepG2 in non-phenol red contained medium while the expressions of CYP1A2 and CYP2E1 in HepG2 were similar in both mediums. Regarding Caco-2, rifampicin significantly induced all CYPs in phenol red contained medium while the induction of CYPs by rifampicin was very low in non-phenol red contained medium. Discussion and Conclusion: Phenol red is one factor affecting the induction of CYPs. Besides, the presence of phenol red leads to the different results depending on the cell lines.
